Time-dependent regulation of perineuronal nets in the cerebellar cortex during abstinence of cocaine-self administration

چکیده

The probability of structural remodeling in brain circuits may be modulated by molecules perineuronal nets (PNNs) that restrict neuronal plasticity to stabilize circuits. Animal research demonstrates addictive drugs can remodel PNNs different regions, including the cerebellum. This study aimed investigate effects short versus extended access cocaine self-administration on PNN expression around Golgi interneurons cerebellar cortex after periods abstinence. After 1 week training (2 h/day), Sprague-Dawley rats self-administered daily for 20 days under (ShA) or (LgA) access. cerebellum was assessed day, 7 days, and 28 forced were immunolabeled using Wisteria floribunda agglutinin (WFA) captured confocal microscopy. WFA intensity increased PNN-bearing neurons over abstinence period a higher proportion more intense formed throughout first month 24 h abstinence, however, we found reduction with ShA as compared naïve animals. When comparing rats, LgA showed consistent upregulation at Our results suggest produces modifications enhance conditions synaptic cortex. These are revealed shortly cessation drug intake but become during protracted group, pointing stabilization drug-induced changes. findings indicate dynamically regulates

in the present study, the effect of e-64 at different concentrations (0.5, 1, 5 and 10 µm) added to (1) the ivm medium on oocyte nuclear maturation and developmental competence of ovine oocytes, and (2) to the ivc medium on embryonic development of ovine embryos were investigated.

The Effect of Heroin Self-Administration on Perineuronal Nets using an Animal Conflict Model of Abstinence and Relapse

Perineuronal Nets (PNNs) are specialized extracellular matrix structures of the brain that are found around specific neurons. PNNs play a role in structural and developmental plasticity, it remains unknown how they are affected by drugs of addiction. In the US, drug use has a very profound impact medically, economically, and socially. We used a conflict model which mimicked human drug seeking b...

Impact of Environmental Enrichment on Perineuronal Nets in the Prefrontal Cortex following Early and Late Abstinence from Sucrose Self-Administration in Rats

Perineuronal nets (PNNs) are aggregates of extracellular matrix that form structures surrounding a subset of GABAergic interneurons. The staining intensity of PNNs appears to be related to plasticity. Environmental enrichment (EE) influences plasticity during adulthood: EE decreases the rewarding effects of drugs of abuse and diminishes both drug- and sucrose-seeking behavior. We determined the...
